β-glucan from Oyster Mushroom (Pleurotus ostreatus) Cultivated in Himalayan Region: Effect of γ-irradiation on Structural, Thermal, Functional, Antioxidant, Antimicrobial, Antiproliferative and Immunomodulatory Properties

ABSTRACT
Background: This study was carried out to evaluate the effect of γ-irradiation on the structural, thermal, functional, antioxidant and antimicrobial, antiproliferative & immunomodulatory properties of β-glucan obtained from the cultivated variety of mushroom, Pleurotus ostreatus. Material and Methods: The structural characterization of the irradiated Pleurotus β-glucan was characterized using ATR- Fourier Transform Infrared and thermal properties were studied using DSC. Results: The functional properties of irradiated Pleurotus β-glucan were largely influenced by the action of gamma irradiation like swelling power decreases with an increase in the irradiation dose while as fat binding capacity, emulsifying properties, foaming properties and bile acid binding capacity shows an increasing trend. All the antioxidant properties like DPPH, reducing power, metal chelating ability of ferrous ion, ABTS & inhibition of lipid per oxidation increased significantly (P ≤ 0.05) in a dose dependent manner. The antibacterial activity of Pleurotus β-glucan also showed an increasing trend with increase in the irradiation dose from 5 to 50 kGy. The antiproliferative activity of Pleurotus β-glucan increased significantly with dose against all the four cell lines and the NO production also showed the similar increasing trend when tested on THP-monocytes.

Conclusion: Irradiated Pleurotus β-glucan proved to be an effective and functional ingredient which can be incorporated in various food formulation in processing units and a potent bioactive component in pharmaceutical companies.
