Antioxidant and Antiproliferative Activities of Hesperidin in MCF-7 Human Breast Cancer Cell Line

ABSTRACT
Background: Hesperidin is a flavanone glycoside consisting of the flavone hesperitin bound to the disaccharide rutinose. The phytochemical hesperidin is mainly found in citrus fruits and has antioxidant and anticarcinogenic activities. MCF-7 human breast cancer cells are used for in-vitro breast cancer studies. Objectives: This study was aimed to examine the effect of hesperidin on MCF-7 cells viability, chromatin condensation, membrane potential, and apoptotic bodies formation. Methods: Determination of MCF-7 cells viability, chromatin condensation, membrane potential, and apoptotic bodies in the presence or absence of hesperidin. Results: The apoptotic activity was evidenced from the chromatin condensation, using DAPI staining. The presence of apoptotic bodies was not observed in MCF-7 cells treated with higher dose of hesperidin (100 μg/mL). The plasma membrane potential of the MCF-7 cells was determined by using Annexin V-FITC staining. Flipping of phosphatidyl serine (PS) was intense in cells treated with positive control, 5-flurouracil, showing strong binding of annexin V with PS .The mitochondrial membrane potential was determined by using JC-1 staining. Significant depolarization of mitochondrial membrane potential was observed in cells treated with hesperidin at higher concentration (100 μg/mL) as evidenced by strong green fluorescence. These results suggest the drop of the inner mitochondrial membrane potential and mitochondrial dysfunction. Hesperidin at lower concentration (50 μg/mL) displayed a blend of green and orange red fluorescence. Conclusion: The dietary derived compound, hesperidin, has a chemotherapeutic activity in MCF-7 cell line. Hesperidin prevents breast cancer development in a dose-dependent manner.
