ABSTRACT
Background: Diabetic nephropathy (DN) is progressively becoming a major challenge for the health care system. Identification of genetic components of diabetic nephropathy is the most important area of diabetes research because elucidation of genes associated with it will lead to better understanding of the disease at molecular level, its related complications, diagnosis, cure, treatment and prevention. Other aim of the research was to study the role of genetic polymorphism of COL4A1(rs565470) gene in increasing susceptibility to DN in North Indian Population. Method: Our study included 50 DN cases and 50 controls. COL4A1 polymorphisms were evaluated using polymerase chain reaction followed by restriction fragment length polymorphism (PCR-RFLP). The genotyping data were compared between cases and controls using chi-square test and logistic regression analysis. Results: We observed a significant difference in cases and controls on comparing the allele frequencies of T and C alleles. The frequency of occurrence of TT genotype and T allele was higher in our cases (52% and 76% respectively) as compared to controls (26% and 54% respectively) (P values were 0.008 and 0.001 respectively) leading to a possibility that they could act as risk allele for prediction of diabetic nephropathy in our set of patients. Conclusion: COL4A1(rs565470) gene polymorphism is associated with the risk of diabetic nephropathy.
